Title : Hepatitis B virus X protein suppresses caveolin - 1 expression in hepatocellular carcinoma through regulating DNA methylation
نویسنده
چکیده
2. The authors suggested that HBx can induce caveolin-1 gene CpG island methylation, based on their in vitro study using a SMMC-7721 cell line. They also found that 28/33 HCC samples were positive for caveoline-1 gene CpG island methylation. How about the adjacent non-tumor tissues? Since HBx may commonly expressed in the non-tumor tissue, has they seen any caveolin-1 CpG island methylation in specimen derived from the HBV positive non-tumor tissues? Also, it is of informative to compare the methylation statues of caveolin-1 gene CpG island between HBV-positive and HBV-negative HCC tissues.

Aberrant DNA methylation in hepatocellular carcinoma tumor suppression (Review)
Aberrant DNA methylation leads to altered gene expression, resulting in cancerous features. Numerous tumor suppressor genes are silenced by DNA methylation during hepatocarcinogenesis. Promoter CpG island hypermethylation is an important mechanism for inactivating tumor suppressor genes in hepatocellular carcinoma (HCC). Hypermethylation of CpG islands in the p16 (INK4a) and p15 (INK4b) promote...
متن کاملExpression of DNA methyltransferase 1 is activated by hepatitis B virus X protein via a regulatory circuit involving the p16INK4a-cyclin D1-CDK 4/6-pRb-E2F1 pathway.
DNA methyltransferase 1 (DNMT1) is responsible for copying DNA methylation patterns to the daughter strands during DNA replication. Its expression is frequently up-regulated in human tumors, including hepatocellular carcinoma, but the mechanism of overexpression and its biological significance remain unclear. Here, we show that hepatitis B virus X protein (HBx) activates DNMT1 expression via a ...
متن کاملEffects of hepatitis C virus NS3 protein on expression of heat shock protein 70 and Glypican3 as the markers of hepatocellular carcinoma
Background and Aims: Hepatitis C virus (HCV) infection is an important risk factor for the development of liver cancer. The HCV NS3 protein plays a key role in the virus life cycle and can affect normal cellular activities, such as cell proliferation, cell death, and cell signaling pathways. Moreover, it may influence malignancy development. Two cellular genes, heat shock protein 70 (HSP70) and...
متن کاملDLEC1 Expression Is Modulated by Epigenetic Modifications in Hepatocelluar Carcinoma Cells: Role of HBx Genotypes
Deleted in Lung and Esophageal Cancer 1 (DLEC1) is a functional tumor suppressor gene (TSG). It has been found to be silenced in a variety of human cancers including hepatocellular carcinoma (HCC). The silencing of DLEC1 can be modulated by epigenetic modifications, such as DNA hypermethylation and histone hypoacetylation. In the case of HCC, hepatitis B virus X protein (HBx) has been implicate...
متن کاملHBx protein‐mediated ATOH1 downregulation suppresses ARID2 expression and promotes hepatocellular carcinoma
Hepatitis B virus X protein plays a crucial role in the pathogenesis of hepatocellular carcinoma. We previously showed that the tumor suppressor ARID2 inhibits hepatoma cell cycle progression and tumor growth. Here, we evaluated whether hepatitis B virus X protein was involved in the modulation of ARID2 expression and hepatocarcinogenesis associated with hepatitis B virus infection. ARID2 expre...
